目的研究斑蝥酸钠维生素B_6诱导MG-63细胞凋亡的作用机制。方法应用不同浓度的斑蝥酸钠维生素B_6作用于MG-63细胞,用MTT比色法检测药物对细胞的增殖抑制作用;反转录聚合酶链反应(RT-PCR)检测存活素信使核糖核酸的表达。结果斑蝥酸钠维生素B_6在抑制MG-63细胞增殖方面有显著作用,其作用具有时间和剂量依赖性,抑制率可高达(68.56±5.21)%。RT-PCR显示可下调survivin基因表达。结论斑蝥酸钠维生素B_6抑制MG-63细胞增殖效果明显,其机制可能与下调survivin基因的表达有关。
Aim To study the mechanism of apoptosis induced by Vitamin C_6 induced by sodium cantharidinate in MG-63 cells. Methods Different concentrations of sodium cantharidinate vitamin B 6 were applied to MG-63 cells. MTT colorimetric assay was used to detect the inhibitory effect of the drugs on cell proliferation. RT-PCR was used to detect the expression of survivin messenger ribonucleic acid expression. Results The sodium cantharidinate vitamin B 6 had a significant effect on inhibiting the proliferation of MG-63 cells. The effect was time-and dose-dependent, with the inhibition rate as high as (68.56 ± 5.21)%. RT-PCR showed down-regulated survivin gene expression. Conclusion Sodium cantharidinate vitamin B 6 can inhibit the proliferation of MG-63 cells obviously. The mechanism may be related to the down-regulation of survivin gene expression.
