目的 研究 5 氮胞苷 (5 aza CR)诱导肺癌细胞系SPC A 1细胞凋亡。方法 MTT法测定SPC A 1细胞毒性 ,荧光显微镜观察细胞DNA的变化 ,流式细胞仪对细胞凋亡分析。结果 5 aza CR对SPC A 1细胞IC5 0在 9.2 7μg/mL ,SPC A 1细胞生长曲线表明 ,随 5 aza CR浓度增高 ,生长率明显下降。细胞凋亡可在 6~ 2 0 μg/ml 5 aza CR处理后 2 4h出现。凋亡细胞主要表现细胞体积缩小 ,核染色质固缩 ,荧光染色增强 ,亚二倍体峰出现。结论 5 aza CR可以诱导SPC A 1细胞发生凋亡。
Aim To investigate the apoptosis of lung cancer cell line SPC A 1 induced by 5-azacytidine (5-aza CR). Methods The cytotoxicity of SPC A 1 was assayed by MTT assay. The changes of DNA were observed by fluorescence microscopy and the apoptosis was analyzed by flow cytometry. Results The IC 50 of 5 aza CR against SPC A 1 cells was 9.2 7 μg / mL. The growth curve of SPC A 1 cells showed that the growth rate of SPC A 1 cells decreased significantly with the increase of 5 aza CR concentration. Apoptosis was observed at 24 h after 20 μg / ml 5 aza CR treatment. Apoptotic cells mainly showed that the cell volume was reduced, the nuclear chromatin was condensed and the fluorescence staining was enhanced, and the sub-diploid peak appeared. Conclusions 5 aza CR can induce apoptosis in SPC A 1 cells.
