目的繁殖和鉴定白介素2(IL-2)基因敲除小鼠。方法将所引进的杂合子小鼠进行饲养并繁殖,提取子代小鼠尾部DNA,用PCR方法检测子代小鼠基因型。结果子代小鼠基因型分别为杂合子(IL-2+/-)、纯合子(IL-2-/-)和野生型(IL-2+/+)。结论 IL-2基因敲除小鼠的繁育和鉴定均获得成功,为进一步研究IL-2缺失如何影响机体免疫功能提供了必要条件。
Objective To breed and identify interleukin 2 (IL-2) knockout mice. Methods The heterozygous mice introduced were bred and propagated. The tail DNA of offspring mice was extracted and the genotypes of offspring mouse were detected by PCR. Results The offspring mouse genotypes were heterozygous (IL-2 +/-), homozygous (IL-2 - / -) and wild-type (IL-2 + / +), respectively. Conclusion The successful breeding and identification of IL-2 knockout mice have provided the necessary conditions for further study on how IL-2 loss affects the immune function.
