目的:建立测定青叶胆中獐牙菜苦苷和异牡荆素含量的高效毛细管电泳分析方法。方法:青叶胆药材粉末经50%甲醇溶液超声提取后,采用未涂层弹性石英毛细管柱(60.2cm×75μm,有效长度50cm)作为分离通道;以含5%甲醇的50mmol·L-l硼砂(pH=9.37)为运行缓冲溶液;分离电压15kV;检测波长236nm;柱温25℃。结果:獐牙菜苦苷和异牡荆素浓度分别在0.1304~1.304mg·mL-l(r=0.9998)和0.0298~0.298mg·mL-l(r=0.9994)范围内呈良好的线性关系;平均回收率(n=6)分别为98.5%和99.3%,RSD均小于2%。结论:本法简便、快速,可用于青叶胆中獐牙菜苦苷和异牡荆素的含量测定。
OBJECTIVE: To establish a high performance capillary electrophoresis method for the determination of swertia glycoside and isovitexin in Bos taurus. Methods: After the powder of Aneurolepidium was extracted by 50% methanol solution, the uncoated elastic quartz capillary column (60.2cm × 75μm, effective length 50cm) was used as the separation channel. With 50 mmol·L of borax containing 5% = 9.37) for running buffer solution; separation voltage 15kV; detection wavelength 236nm; column temperature 25 ℃. Results: The concentrations of swertia glycoside and isovitexin were linear in the range of 0.1304-1.304 mg · mL-1 (r = 0.9998) and 0.0298-0.298 mg · mL-1 (r = 0.9994), respectively. The average recovery (n = 6) was 98.5% and 99.3% respectively, with RSDs less than 2%. Conclusion: This method is simple, rapid and can be used for the determination of swertia glycoside and isovitexin in Aloe vera.
