本实验用Wistar大鼠建立三种脑损伤模型,即脑顶叶皮层针刺损伤、全脑缺血/再灌注和急性高原(5000米)缺氧,应用胶质原纤维酸性蛋白(GFAP)单克隆抗体免疫组化方法,观察了伤后1天,3天,5天,7天,10天,14天,21天皮层、海马及室周区星形胶质细胞的形态学变化。结果如下:(1)针刺脑损伤组:术后5-7天,伤侧海马CAl、CA。区及室周区可见星形胶质细胞染色加深,胞体增大,数目增加,伤道底部GFAP阳性纤维长短不一;至14天左右阳性细胞突起增长增粗,数目进一步增加,伤道底部阳性纤维横向排列,多垂直于伤道。(2)全脑缺血/再灌注组:术后1天即可见海马CA;、CA。区星形胶质细胞染色增强,胞体增大;术后3-5天,皮层、海马CAl、CA。区及室周区星形胶质细胞脑体增大,突起增粮、增长,染色加深,细胞数目产多,海马齿状回处GFAP阳性纤维较正常增多增长,且呈辐射状排列:水后14-ZI天,仍见上述反应,但无进一步改变。(3)急性高原缺氧组;1—5天,星形胶质细胞未见明显形态学变化;10—14天,可见有轻微的GFAP阳性细胞数目增多及少数细胞胞体增大。

(3) Acute plateau hypoxia group; 1-5 days, no obvious morphological changes of astrocytes; 10-14 days, showing a slight increase in the number of GFAP positive cells and a small number of cells increased.
