目的 比较弗氏完全佐剂(FCA)与单磷脂A(MPL)的免疫增强作用,为提高重组SjG-Sj32(rSjGST-sJ32)的疫苗效果提供依据。方法 rSjGST-Sj32加弗氏完全佐剂(rSjGST-Sj32+FCA)或单磷脂A(rSjGST-Sj32+MPL)免疫小鼠3次,ELISA法检测抗体水平,用减虫率及减卵率表示保护性作用。结果 与PBS对照组相比,rSjGST-Sj32加FCA或MPL免疫BALB/c小鼠减虫率分别为48.8%(P<0.01)和44.1%(P<0.01);每克肝卵(LEPG)减少率分别为59.4%(P<0.01)和46.2%(P<0.01);rSjGST+Sj32+FCA组抗体滴度达1:204800(P<0.05),而rSjGST-Sj32+MPL组抗体滴度为1:51200(P<0.05)。结论 FCA和MPL均可以增强rSjGST-Sj32免疫小鼠的抗感染保护性免疫力;FCA也可增强rSjGST-Sj32诱导小鼠抗生殖免疫,而MPL似不具明显增强rSjGST-Sj32的抗生殖免疫作用。佐剂MPL在提高免疫小鼠体液免疫应答方面稍逊于经典佐剂FCA。“,”Aim To compare the immunoenhancing effects between two adjuvants, Freund’s complete adjuvant (FCA) and monophosphoryl lipid A (MPL), in vaccination of mice with recombinant fusion protein SjGST-SJ32 (rSjGST-Sj32) .Methods Mice were immunized three times with rSjGST-Sj32 plus FCA or MPL.Levels of specific antibodies were detected by ELISA.After challenge, the mice were perfused, the wonms and the eggs which collected from the livers of mice were counted. Results significant levels of specific antibody were induced in mice immunized wth rSjGST-Sj32 plus either FCA or MPL. As compared with the control group, the worm reduction rates were 49% and 44% respectively; the liver egg pergram (LEPG) were 59% and 46% respectively, and the liver egg perfemale (LEPF) reduction rates were 35% and 19% respectively. Conclusion The results show that FCA or MPL plus rSjGST-Sj32 could significantly induce protective immunity, however, the former induced significantly anti-fecundity immumity, but the latter had not such an effect. Adjuvant MPL was not as good as FCA in enhancing humoral immune response induced by rSjGST-Sj32 in mice.
