为探讨NADH对PC12细胞化学药物损伤后的修复作用 ,采用细胞实验检测NADH对顺铂、阿霉素、鱼藤酮对肾上腺髓质嗜铬细胞瘤PC12细胞系细胞毒性作用的影响 ,并运用流式细胞仪对细胞周期蛋白、G蛋白和转谷酰胺酶Ⅱ含量进行分析。结果发现 ,NADH能够明显抑制化学药物顺铂、阿霉素、鱼藤酮对PC12细胞的毒性作用 ,上调细胞损伤后周期蛋白A和B1表达及下调周期蛋白D1表达 ;上调G蛋白和转谷酰胺酶Ⅱ表达。提示NADH抑制和修复化学药物对PC12细胞的损伤与细胞周期蛋白、G蛋白和转谷酰胺酶Ⅱ调节有关
To investigate the repair effect of NADH on PC12 cells after chemical injury, the effect of NADH on the cytotoxicity of cisplatin, doxorubicin and rotenone on PC12 cell line of adrenal medullary pheochromocytoma was tested by cell assay. Flow cytometry The contents of cyclin, G protein and transglutaminase II were analyzed. The results showed that NADH could significantly inhibit the toxic effects of chemotherapeutic agents cisplatin, doxorubicin and rotenone on PC12 cells, upregulate cyclin A and B1 and downregulate cyclin D1, upregulate G protein and transglutaminase Ⅱ expression. These results suggest that NADH suppresses and repairs the injury of PC12 cells induced by chemical drugs and is related to the regulation of cyclin, G protein and transglutaminase II
