目的:观察中药复方肠复康(CFK)动物血清对HUVEC血管形成及Gli 1、VEGF表达的影响。方法:采用双室联合培养人脐静脉内皮细胞(HUVEC)和人结肠癌(Lovo)细胞,观察CFK含药血清对HUVEC血管形成的影响,并观察Gli 1及VEGF表达的变化。结果:CFK含药血清作用后,HUVEC增殖活性明显降低,其成管指数明显降低,Gli 1、VEGF表达明显减少,与对照组比较均有显著性差异(P<0.05或P<0.01)。结论:CFK具有抑制大肠癌血管生成作用。其机制与其抑制Hedgehog-Gli1信号通路激活,下调VEGF表达而发挥抗肿瘤血管形成作用有关。
Objective: To observe the effects of serum of traditional Chinese medicine compound Changfukang (CFK) on angiogenesis and the expression of Gli 1 and VEGF in HUVECs. Methods: Human umbilical vein endothelial cells (HUVECs) and human colon cancer cells (Lovo) were co-cultured in vitro. The effects of CFK-containing serum on the formation of HUVEC were observed. The changes of Gli 1 and VEGF were observed. Results: The proliferation of HUVEC significantly decreased after treated with CFK. The tube formation index was significantly decreased, while the expression of Gli 1 and VEGF was significantly decreased (P <0.05 or P <0.01). Conclusion: CFK can inhibit the angiogenesis of colorectal cancer. Its mechanism and its inhibition of Hedgehog-Gli1 signaling pathway activation, downregulation of VEGF expression and exert anti-tumor angiogenesis.
