目的:研究滋阴方二至丸对二甲基苯蒽(DMBA)诱发性乳腺癌大鼠肿瘤组织中血管内皮生长因子(VEGF)、基质金属蛋白酶-9(MMP-9)表达的影响。方法:60只雌性SD大鼠随机分为空白对照组、DMBA模型组、二至丸高、中、低(6.4,3.2,1.6 g.kg-1)剂量组和三苯氧胺(TAM)组。除空白对照组外,每只大鼠均以致癌剂DMBA诱导(剂量100 mg.kg-1,2次,间隔1周)乳腺癌。诱导第2天开始给予药物干预,每周触诊所有大鼠乳腺肿瘤发生情况并记录体重变化;15周后处死动物,统计各组大鼠乳腺肿瘤的发病率,并采用SP免疫组化法检测各组动物肿瘤组织中VEGF,MMP-9的表达情况。结果:与DMBA模型组比较,二至丸中剂量组和TAM组大鼠乳腺肿瘤平均潜伏期延长(P<0.05),平均肿瘤直径、平均肿瘤体积、平均瘤重显著降低(P<0.05);二至丸中、高剂量组VEGF和MMP-9强阳性率显著性降低(P<0.05),且VEGF与MMP-9的表达呈正相关。结论:滋阴方药二至丸可能通过干预乳腺癌组织中VEGF和MMP-9的表达,抑制乳腺癌生长恶化。
Objective: To study the effect of Ziyin Fang Erzhi Pill on the expression of vascular endothelial growth factor (VEGF) and matrix metalloproteinase-9 (MMP-9) in tumor tissues induced by dimethylbenzyl anthracene (DMBA) in rats. Methods: Sixty female Sprague-Dawley rats were randomly divided into blank control group, DMBA model group, Erzhi pill high, low dose (6.4,3.2,1.6 g.kg-1) dose group and tamoxifen (TAM) group. Except for the blank control group, each rat was induced with DMBA, a carcinogen, at doses of 100 mg.kg-1, 2 times at 1 week intervals. On the second day after the initiation of induction, drug intervention was given. All the rats were palpated and the weight of the mammary gland was palpated weekly. After 15 weeks, the animals were sacrificed and the incidence of breast tumors was calculated. The SP immunohistochemical method The expression of VEGF and MMP-9 in each group of animal tumor tissues. Results: Compared with DMBA model group, the average latency of breast cancer in Erzhiwan middle dose group and TAM group was longer (P <0.05), mean tumor diameter, mean tumor volume and mean tumor weight were significantly lower (P <0.05) However, the positive rates of VEGF and MMP-9 in high dose group were significantly lower (P <0.05), and the expression of VEGF and MMP-9 were positively correlated. Conclusion: Ziyin Prescription, Erzhi Pill, may inhibit the growth of breast cancer by interfering with the expression of VEGF and MMP-9 in breast cancer.
