目的观察植物抗菌液(PAS)的抗炎作用并探讨其机制。方法分别用巴豆油和2,4-二硝基氯苯制备小鼠耳肿胀和耳接触性皮炎模型;检测小鼠耳廓肿胀度和血清中肿瘤坏死因子(TNF-α)的水平;观察小鼠致炎耳的结痂情况,测定致炎耳的厚度增加值和与正常耳的重量差;测定血清中白介素4(IL-4)和白介素12(IL-12)的含量。结果 PAS组小鼠的耳廓肿胀度和血清中TNF-α水平低于模型组的;模型组小鼠的耳结痂数多于PAS组的;PAS组的耳廓厚度增加值和与正常耳的重量差均明显低于模型组的;血清中IL-12的水平高于模型组的,IL-4的水平低于模型组的。结论 PAS能有效抑制巴豆油引起的小鼠耳肿胀,其机制可能和降低血清中TNF-α的水平有关;PAS还能有效抑制小鼠耳廓的接触性皮炎,其机制可能和升高血清中IL-12的水平和降低IL-4的水平有关。
Objective To observe the anti-inflammatory effect of plant antibacterial solution (PAS) and to explore its mechanism. Methods The models of ear swelling and ear contact dermatitis in rats were made by using croton oil and 2,4-dinitrochlorobenzene respectively. The auricle swelling degree and serum level of tumor necrosis factor (TNF-α) were measured. Rat sclerosing ear scabs were measured, the thickness of the ear caused by the increase in the value and the normal ear weight difference; determination of serum interleukin 4 (IL-4) and interleukin 12 (IL-12) content. Results The degree of auricle swelling and serum TNF-α in PAS group were lower than those in model group. The number of ear scab in model group was more than that of PAS group. The weight difference was significantly lower than the model group; serum IL-12 levels higher than the model group, IL-4 levels lower than the model group. Conclusions PAS can effectively inhibit ear swelling in mice induced by croton oil, and its mechanism may be related to the decrease of TNF-α level in serum. PAS can also inhibit contact dermatitis in mouse auricles. The mechanism may be related to increasing serum levels of IL-12 levels and reduce the level of IL-4.
