目的观察anti-miR-155反义寡核苷酸(AMOs)对人胰腺癌细胞株SW1990增殖和凋亡的影响。方法将SW1990细胞分为空白组、阴性对照组和AMOs处理组。阴性对照组将阴性对照链转染入SW1990细胞株中,AMOs处理组采用AMOs抑制SW1990细胞内miR-155的活性;CCK-8法测定细胞增殖抑制率,流式细胞术测定细胞周期及凋亡。结果与空白组及阴性对照组相比,AMOs显著抑制SW1990细胞的增殖(P<0.05),100nmol/L AMOs的抑制率达36.51%,使G0/G1细胞比例显著增加,G2/M期细胞比例显著减少(P<0.05),AMOs处理过的SW1990细胞早期凋亡率明显增高(P<0.05)。结论 AMOs通过抑制SW1990细胞内高水平表达miR-155的活性,显著抑制SW1990细胞的增殖。
Objective To investigate the effects of anti-miR-155 antisense oligonucleotides (AMOs) on the proliferation and apoptosis of human pancreatic cancer cell line SW1990. Methods SW1990 cells were divided into blank group, negative control group and AMOs treatment group. Negative control group transfected negative control strand into SW1990 cell line, AMOs treatment group using AMOs inhibit SW1990 cell miR-155 activity; CCK-8 method to determine the inhibition rate of cell proliferation, flow cytometry determination of cell cycle and apoptosis . Results Compared with the blank control group and the negative control group, AMOs significantly inhibited the proliferation of SW1990 cells (P <0.05), and the inhibitory rate of 100 nmol / L AMOs was 36.51%. The proportion of G0 / G1 cells was significantly increased. (P <0.05). The early apoptotic rate of SW1990 cells treated with AMOs was significantly increased (P <0.05). Conclusion AMOs can significantly inhibit the proliferation of SW1990 cells by inhibiting the high level expression of miR-155 in SW1990 cells.
