自从建立化学结构理论以来,研究结构与反应性能的依賴关系,一直是有机化学中的首要理论问题。在给定的环境条件下,分子的性质取决于分子的电子层结构或分子的精细结构;而后者又是以分子的骨架结构,即分子中原子或原子核的连接顺序以及它们的室间配置为基础。经典有机化学,主要地凭借化学研究的方法,解决了确定分子骨架结构的问題;从而建立了許多关于分子结构与反应性能的概念和规律。
