目的 探讨雷公藤T4 单体 (简称T4 )、细胞因子对人成纤维细胞增殖作用的影响 ,从而为青光眼滤过手术成功率的提高及与瘢痕形成有关的眼疾病的防治提供理论基础和有效措施。方法 人成纤维细胞常规传代培养 ,分别加入 5 氟尿嘧啶 (5 fluo rouracil,5 Fu )、T4 、干扰素 (interferonalpha 2b ,IFNα 2b)、白细胞介素 2 (interleukin 2 ,IL 2 )进行药物实验 ,并用四唑蓝 (MTT )比色法检测。结果 T4 、5 Fu对人成纤维细胞增殖有抑制作用 ,IFNα 2b、IL 2对人成纤维细胞增殖有促进作用 ;T4 与IL 2联合应用对人成纤维细胞增殖无影响 ,T+4IFNα 2b对成纤维细胞增殖有抑制作用。结论 雷公藤T4 单体可能是青光眼滤过术后抗瘢痕形成的有效药物。T4与IFNα 2b联合应用可能是青光眼滤过术后抗瘢痕形成的有效方法。
Objective To investigate the effects of Tripterygium wilfordii T4 monomer (T4) and cytokines on the proliferation of human fibroblasts, so as to provide theoretical basis and effective measures for improving the success rate of glaucoma filtration surgery and prevention and treatment of eye diseases related to scar formation . Methods Human fibroblasts were subcultured by conventional methods. Drugs such as 5-fluorouracil (5 Fu), interferon-α 2b (IFNα 2b) and interleukin 2 (IL 2) Tetrazolium blue (MTT) colorimetric assay. Results T4 and 5 Fu could inhibit the proliferation of human fibroblasts. IFNα 2b and IL 2 could promote the proliferation of human fibroblasts. The combination of T4 and IL 2 had no effect on the proliferation of human fibroblasts. T + 4 IFNα 2b Fibroblast proliferation inhibited. Conclusion Tripterygium T4 monomer may be an effective anti-scarring agent for glaucoma filtration. The combined use of T4 and IFNα 2b may be an effective method of anti-scar formation after glaucoma filtration surgery.
