目的探讨Notch1胞内段(NIC)在非角化性鼻咽癌组织和细胞株中的表达。方法采用反转录聚合酶链反应(RT-PCR)及免疫组化染色方法对鼻咽癌标本及不同分化程度鼻咽癌细胞株进行检测。结果 RT-PCR检测显示不同鼻咽癌细胞株中NIC基因均有表达,但分化程度高的CNE1细胞株表达最明显;同期进行的免疫组化染色结果与之相似;对鼻咽癌标本的免疫组化染色结果显示:鼻咽癌组织中NIC的表达高于鼻咽慢性黏膜炎症组织(P=0.005),而且NIC的表达强度与细胞分化程度密切相关(P<0.05),与患者性别、年龄无相关性(P>0.05)。另外,在未分化型鼻咽癌中,癌巢内几乎无阳性表达,淋巴细胞阳性表达多;而在分化型鼻咽癌中癌细胞阳性表达明显,可见于胞膜、胞质及胞核。结论 Notch信号系统参与鼻咽癌的发生,其中Notch1的表达与鼻咽癌的分化程度相关。
Objective To investigate the expression of Notch1 intracellular domain (NIC) in non-keratinizing nasopharyngeal carcinoma tissues and cell lines. Methods Reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry were used to detect nasopharyngeal carcinoma specimens and nasopharyngeal carcinoma cell lines with different degrees of differentiation. Results RT-PCR showed that the expression of NIC gene in different nasopharyngeal carcinoma cell lines was highly expressed, but the most highly differentiated CNE1 cell line was the most obvious. The immunohistochemical staining results were similar in the same period; The results of histochemical staining showed that the expression of NIC in nasopharyngeal carcinoma tissues was higher than that in chronic nasopharyngeal mucosa tissues (P = 0.005), and the expression intensity of NIC was closely related to the degree of cell differentiation (P <0.05) No correlation (P> 0.05). In addition, in undifferentiated nasopharyngeal carcinoma, almost no positive expression of cancer nest, lymphocyte positive expression and more; in differentiated NPC positive expression of cancer cells can be found in the membrane, cytoplasm and nucleus. Conclusion The Notch signaling system is involved in the occurrence of nasopharyngeal carcinoma. The expression of Notch1 is correlated with the differentiation of nasopharyngeal carcinoma.
