目的体外观察脐带华通胶间充质干细胞(UCWJMSCs)对急性再生障碍性贫血(AAA)患者T淋巴细胞(TLC)亚群的调节作用。方法收集21例AAA患者和10例健康志愿者外周血,制备单个核细胞(MNCs)悬液,UC-WJMSCs和MNCs以一定比例混合共育一定时间后,采用流式细胞仪检测TLC亚群并进行统计分析。结果 AAA患者TLC亚群呈Th1、Tc1极化状态。混合培养后,健康人的TLC亚群未见明显变化(P>0.05);AAA患者的TLC亚群除Th2与Tc2无明显变化外,CD3+、CD8+、CD2+8、Th1和Tc1亚群比例明显降低,而CD4+、CD4+/CD8+、CD5+6、CD2+5和CD1+03亚群比例明显升高(P<0.05)。调节作用24 h就已产生7,2 h时达高峰;调节作用随UCWJMSCs浓度的增高而增强。结论 UCWJMSCs对AAA患者的TLC亚群有明显调节作用,不同个体来源的UCWJMSCs调节作用无区别。
Objective To observe the regulatory effect of umbilical cord blood mesenchymal stem cells (UCWJMSCs) on T lymphocyte subsets in patients with acute aplastic anemia (AAA) in vitro. Methods Peripheral blood was collected from 21 patients with AAA and 10 healthy volunteers. Mononuclear cells (MNCs) suspension were prepared. UC-WJMSCs and MNCs were co-cultured for a certain period of time. Flow cytometry was used to detect the subpopulations of TLC conduct statistical analysis. Results TLC subpopulations of patients with AAA showed Th1, Tc1 polarization state. There was no significant change in the TLC subpopulation of healthy people (P> 0.05) after mixed culture. The proportion of CD3 +, CD8 +, CD2 + 8, Th1 and Tc1 subsets in TLC subgroups of AAA patients was significantly lower than that of Tc2 (P <0.05), while the proportion of CD4 +, CD4 + / CD8 +, CD5 + 6, CD2 + 5 and CD1 + 03 subsets was significantly increased. The regulatory effect reached the peak at 7 h and reached the peak at 24 h. The regulatory effect increased with the increase of UCWJMSCs concentration. Conclusions UCWJMSCs can obviously regulate the subpopulation of AAA in patients with AAA, and there is no difference in the regulation of UCWJMSCs derived from different individuals.
