目的:探讨不同抗原修复对A549肿瘤组织内Ki-67蛋白检测结果的影响。方法:采用酶修复法、柠檬酸微波修复法、柠檬酸高压修复法、Tris/EDTA微波修复法、Tris/EDTA高压修复法等对肿瘤组织的石蜡切片进行修复,采用二步法进行免疫组织化学染色。结果:肿瘤组织的核蛋白Ki-67经柠檬酸微波修复后,染色效果较其他修复方法明显增强,且定位佳,对比度清晰。结论:在核蛋白Ki-67免疫组织化学染色中,柠檬酸微波修复法优于其他抗原修复方法。
Objective: To investigate the effect of different antigens on the detection of Ki-67 in A549 tumor tissues. Methods: Paraffin sections of tumor tissues were repaired by enzymatic repair method, citric acid microwave repair method, citric acid high pressure repair method, Tris / EDTA microwave repair method and Tris / EDTA high pressure repair method, and immunohistochemistry dyeing. Results: The nuclear protein Ki-67 in tumor tissue was significantly improved after microwave repair by citric acid compared with other methods, and its localization was good and the contrast was clear. CONCLUSION: In the nuclear protein Ki-67 immunohistochemical staining, citric acid microwave repair method is superior to other antigen retrieval methods.
