目的:观察天麻素对长春新碱诱导大鼠神经病理性疼痛的影响。方法:用长春新碱125μg/kg隔日腹腔注射制作大鼠化疗痛动物模型,腹腔注射天麻素治疗;用电子Von Frey测痛仪测定大鼠机械痛阈值,电子压痛仪测定大鼠压痛痛阈值,电子热板仪测定大鼠热刺激痛阈值。结果:腹腔注射长春新碱在给药第10天与对照组比较三种刺激痛阈值均有显著性降低(P<0.01),化疗痛模型成功建立之后,加用天麻素腹腔注射1~2周,不同剂量天麻素组(30mg/kg、60mg/kg、120mg/kg)的三种刺激痛阈值与模型组比较均有不同程度的提高,且呈剂量依赖性,以大剂量天麻素的作用最明显(P<0.01),且天麻素治疗1周与治疗2周的痛阈值无差异。结论:天麻素对长春新碱诱导的大鼠痛觉过敏有抑制作用;持续给药该作用可以维持,有利于延长长春新碱抗肿瘤的疗程,以提高其抗肿瘤疗效。
Objective: To observe the effects of gastrodin on vincristine-induced neuropathic pain in rats. Methods: The rat model of chemotherapy-induced pain was established by intraperitoneal injection of vincristine 125μg / kg every other day. The rats were injected intraperitoneally with gastrodin. The Von Frey apparatus was used to measure the mechanical pain threshold. The electronic pressure- Determination of rat thermal stimulus pain threshold by electronic hot plate. Results: The pain thresholds of three kinds of stimulation of vinblastine were significantly decreased (P <0.01) on the 10th day after administration compared with the control group. After the successful establishment of the model of chemotherapy and pain, 1 ~ 2 weeks of intraperitoneal injection of gastrodin , And the thresholds of three stimuli of gastrodin group (30mg / kg, 60mg / kg, 120mg / kg) were increased in different degrees compared with the model group in a dose-dependent manner. The effect of high-dose gastrodin (P <0.01), and there was no difference in the pain threshold between gastrodin treatment for 1 week and treatment for 2 weeks. Conclusion: Gastrodin can inhibit vomitamia-induced hyperalgesia in rats. Sustained administration of the gastrodin can maintain the anti-tumor effect of vincristine and prolong the anti-tumor activity of vincristine.
