目的:针对传统回流提取易导致有效成分降解的弊端,研究破壁浸提精制冠心颗粒的最优工艺。方法:以芍药苷、阿魏酸与丹酚酸B含量的综合评分为评价指标,采用单因素结合正交试验考察溶媒用量、浸提次数、浸提温度与浸提时间对提取工艺的影响。结果:最佳提取工艺为药材粉碎至d0.5为14μm左右,加入8倍量水,在60℃条件下浸提3次,每次10 min。结论:该优选工艺稳定可行,能在提取环节显著增加有效成分的提取量,可代替传统的回流工艺。
Objective: In view of the drawbacks that the traditional reflux extraction can easily lead to the degradation of active ingredients, the optimum process of broken-leaching refined Guanxin granules is studied. Methods: Taking the comprehensive score of paeoniflorin, ferulic acid and salvianolic acid B content as evaluation index, the effects of solvent dosage, extraction times, extraction temperature and extraction time on the extraction process were investigated by single factor and orthogonal experiment. Results: The optimum extraction process was pulverization of medicinal materials to d0.5 of about 14μm, adding 8 times the amount of water, leaching three times at 60 ℃ for 10 minutes. Conclusion: The optimal process is stable and feasible, which can significantly increase the extraction of active ingredients in the extraction process, which can replace the traditional reflux process.
