目的通过观察青藤碱联合卡铂对宫颈癌患者癌细胞抑制的影响,探讨青藤碱联合卡铂治疗方案的临床价值。方法体外培养人宫颈癌Hela细胞,取对数生长期细胞分为4组,分别为对照组(6孔,只加培养液)、青藤碱组(6孔,只加2.5 mmol/L青藤碱)、卡铂组(6孔,只加40μg/ml卡铂)、联合用药组(分为3个亚组,每个亚组6孔,分别加0.625 mmol/L青藤碱+40μg/ml卡铂、1.25 mmol/L青藤碱+40μg/ml卡铂及2.5 mmol/L+青藤碱40μg/ml卡铂),处理24、48及72 h后,分别以MTT法检测细胞增殖变化,并观测48 h细胞周期变化及不同浓度的青藤碱对细胞凋亡的影响。结果各组细胞增殖情况均受到显著抑制,高浓度联合用药组宫颈癌细胞抑制率显著高于其他各组,青藤碱联合卡铂的抑制作用与时间和浓度呈正比,差异具有统计学意义(P<0.05)。与对照组相比,青藤碱组G1期细胞明显增多,S期细胞明显减少,卡铂组G1期细胞明显减少,S期细胞明显增多;联合用药组的G1期细胞与S期细胞位于两组之间。高浓度联合用药组凋亡率(75.58%)显著高于其他组(53.33%),且随着时间增加而升高,差异具有统计学意义(P<0.05)。结论青藤碱联合卡铂能够影响细胞周期分布,促进细胞凋亡,从而显著抑制宫颈癌细胞增殖,是宫颈癌较为有效的辅助治疗手段。
Objective To observe the effect of sinomenine combined with carboplatin on the inhibition of cancer cells in patients with cervical cancer and to explore the clinical value of sinomenine combined with carboplatin in the treatment. Conclusion Sinomenine combined with carboplatin can affect the cell cycle distribution, promote apoptosis, and thus significantly inhibit cervical cancer cell proliferation, cervical cancer is more effective adjuvant therapy.
