目的 观察缺血预处理对沙土鼠脑缺血再灌注后脑组织线粒体功能的影响。方法 用沙土鼠双侧颈总动脉结扎制成全脑缺血模型 (缺血 2 0分钟 ,再灌注 30分钟 )。动物随机分为预缺血组、模型组和假手术组。预缺血组给予两次 (各 2分钟 )缺血预处理 (间隔 48小时 )。用氧电极法测定呼吸功能和呼吸链的氧化酶(NADH氧化酶、琥珀酸氧化酶、细胞色素 C氧化酶 )活性。结果 缺血模型组动物的呼吸控制率、磷氧比及氧化磷酸化效率均较假手术组有不同程度的降低 ,3种氧化酶的活性亦较假手术组明显降低。而预缺血组的这种改变不明显 ,各测定值接近假手术组 ,但与模型组的差异显著。结论 缺血预处理可以减轻缺血再灌注后脑线粒体功能的损伤。
Objective To observe the effect of ischemic preconditioning on the mitochondrial function of brain tissue in gerbils after cerebral ischemia reperfusion. Methods The model of global cerebral ischemia was established by ligating the common carotid arteries of gerbils (ischemic 20 minutes, reperfusion 30 minutes). Animals were randomly divided into pre-ischemic group, model group and sham operation group. The ischemic group was given ischemic preconditioning (48 hours apart) twice (2 minutes each). The respiratory function and respiratory chain oxidase (NADH oxidase, succinate oxidase, cytochrome C oxidase) activity were measured by the oxygen electrode method. Results The respiratory control rate, P / O ratio and oxidative phosphorylation efficiency of the animals in the ischemic model group were all lower than those in the sham operation group. The activities of the three oxidases were significantly lower than those in the sham operation group. The pre-ischemic group of this change is not obvious, the measured values close to the sham group, but significant differences with the model group. Conclusion Ischemic preconditioning can reduce the damage of brain mitochondrial function after ischemia-reperfusion.
