目的:制备抗小鼠白细胞介素2受体(IL-2R)单克隆抗体(McAb)。方法:利用杂交瘤技术获得杂交瘤细胞株,并用细胞ELISA方法筛选阳性克隆。结果:得到3株分泌抗小鼠IL-2RMcAb的杂交瘤细胞株,其中8C8对ConA刺激淋转有抑制作用,且sIL-2R可抑制8C8与IL-2Rα阳性细胞的结合。结论:8C8是抗小鼠IL-2Rα抗体。
Objective: To prepare anti-mouse interleukin-2 receptor monoclonal antibody (McAb). Methods: The hybridoma cell lines were obtained by hybridoma technique and the positive clones were screened by ELISA. RESULTS: Three hybridoma cell lines secreting anti-mouse IL-2RMcAb were obtained. 8C8 inhibited the lymph node metastasis of ConA and sIL-2R inhibited the binding of 8C8 to IL-2Rα-positive cells. Conclusion: 8C8 is an anti-mouse IL-2Rα antibody.
