目的评价喷鼻三价流感裂解疫苗在小鼠体内产生的免疫原性及免疫保护性。方法选用H1N1、H3N2、B型3种裂解抗原联合后,与佐剂壳聚糖季铵盐(HTCC)水凝胶等体积混合,制备三价流感裂解疫苗。经喷鼻免疫BALB/c小鼠,免疫剂量为每种单价抗原2μg/只,第0及14天各免疫1次,于末次免疫后第14天制备小鼠血清及鼻、肺、阴道灌洗液。血凝抑制法检测小鼠血清的血凝抑制抗体(HI)效价;ELISA法检测小鼠血清中Ig G及其亚型抗体效价和小鼠鼻、肺、阴道灌洗液中sIgA效价;末次免疫14 d,用H1N1流感病毒进行攻毒,观察小鼠体重变化及死亡率。结果经喷鼻免疫三价流感裂解疫苗的小鼠血清可产生较高效价的HI抗体,IgG抗体亚型以IgG1与IgG2a为主,小鼠鼻、肺、阴道灌洗液中均可检测到特异性sIgA。攻毒后小鼠体重变化较小,14 d后全部存活。结论经喷鼻免疫三价流感裂解疫苗可激发小鼠产生体液、细胞及黏膜免疫反应,具有良好的免疫保护性。
Objective To evaluate the immunogenicity and immunoprotective effect of nasal trivalent flu splitting vaccine in mice. Methods Three H1N1, H3N2 and B type cleavage antigens were selected and mixed with equal volume of adjuvant chitosan quaternary ammonium salt (HTCC) hydrogel to prepare trivalent flu cleaving vaccine. BALB / c mice were immunized by nasal spray at the immunization dose of 2μg / unit of each antigen and immunized on the 0th and the 14th day respectively. Serum and nasal, pulmonary and vaginal lavage were prepared on the 14th day after the last immunization liquid. The hemagglutination inhibition method was used to detect the hemagglutination inhibition antibody (HI) titer of mouse sera. The titer of Ig G and its subtype antibody in mouse sera and the sIgA titer in nasal, pulmonary and vaginal lavage fluid of mice At 14 days after the last immunization, the mice were challenged with the H1N1 influenza virus to observe the changes of body weight and mortality. Results The mice immunized with trivalent flu vaccine by nasal immunization produced high titer anti-HI antibody. The IgG antibody subtype was mainly IgG1 and IgG2a. The specific nasal, pulmonary and vaginal lavage fluid was detected in mice Sex sIgA. After challenge, the body weight of mice changed little, all survived after 14 days. Conclusion The nasal immunization trivalent flu cleavage vaccine can stimulate mice to produce humoral, cellular and mucosal immune response, has good immune protection.
