目的:观察丹红化瘀口服液对糖尿病肾病(DN)大鼠的影响。方法:选取8周龄SPF级健康雄性Wistar大鼠48只,随机分为正常对照组、模型组、胰岛素(4.0 U/kg)阳性对照组及丹红化瘀口服液(3.2 m L/kg)组,每组12只,采用链脲佐菌素(STZ)建立DN大鼠模型(血糖水平≥16.7 mmol/L、尿糖水平++++以上),对比各组大鼠给药12 w后血糖、血脂、NO、SOD水平及肾功能;采用逆转录-聚合酶链反应(RT-PCR)技术检测各组大鼠肾脏组织转化生长因子β1(TGF-β1)、p38MAPK、Caspase-3 mRNA的表达。结果:胰岛素组和丹红化瘀口服液组大鼠血糖、TG、TC、BUN、Scr、24h尿蛋白水平显著低于模型组,NO、SOD水平显著高于模型组(P<0.05),肾脏组织TGF-β1、p38MAPK、Caspase-3 mRNA表达显著低于模型组(P<0.05)。结论:丹红化瘀口服液对DN大鼠具有一定的肾脏保护作用,其作用机制可能与调节肾脏TGF-β1、p38MAPK及Caspase-3 mRNA表达水平有关。
Objective: To observe the effect of Danhong Huayu oral liquid on diabetic nephropathy (DN) rats. Methods: Forty-eight healthy male Wistar rats of 8 weeks old were randomly divided into normal control group, model group, 4.0 U / kg insulin positive control group and Danhong Huayu oral solution (3.2 m L / kg) (12 rats in each group). DN rats were induced by streptozotocin (STZ) (blood glucose level≥16.7 mmol / L and urine glucose level ++++) Blood glucose, blood lipids, NO, SOD and renal function were measured. The expression of TGF-β1, p38MAPK and Caspase-3 mRNA in renal tissue was detected by reverse transcription polymerase chain reaction expression. Results: The levels of blood glucose, TG, TC, BUN, Scr and 24h urinary protein in insulin group and Danhonghuayu oral liquid group were significantly lower than those in model group, and the levels of NO and SOD were significantly higher than those in model group (P <0.05) The expression of TGF-β1, p38MAPK and Caspase-3 mRNA in the model group was significantly lower than that in the model group (P <0.05). CONCLUSION: Danhong Huayu oral liquid has a protective effect on kidney of DN rats, and its mechanism may be related to the regulation of renal TGF-β1, p38MAPK and Caspase-3 mRNA expression.
