目的探讨Twist和HIF-1α在鼻咽癌组织中表达及其相关性。方法应用RT-PCR及SABC免疫组化法检测50例鼻咽癌组织Twist、HIF-1α的mRNA及蛋白表达情况。结果鼻咽癌组织中不同患者年龄、肿瘤大小分组间Twist及HIF-1α的mRNA和蛋白表达差异无统计学意义;不同淋巴结转移、临床分期和病理分级分组间Twist及HIF-1α的mRNA和蛋白表达差异具有统计学意义。鼻咽癌组织中Twist的mRNA及蛋白表达间、HIF-1α的mRNA及蛋白表达间、Tw ist和HIF-1α的蛋白表达间、Tw ist和HIF-1α的mRNA表达间均存在正相关关系。结论鼻咽癌组织中Twist、HIF-1α均高表达,且两者间具有相关性。HIF-1α蛋白可能通过上调Twist表达成为其促进鼻咽癌浸润、转移的途径之一。
Objective To investigate the expression of Twist and HIF-1α in nasopharyngeal carcinoma and their correlation. Methods The mRNA and protein expressions of Twist and HIF-1α in 50 cases of nasopharyngeal carcinoma were detected by RT-PCR and SABC immunohistochemistry. Results There was no significant difference in the mRNA and protein expressions of Twist and HIF-1α among the different age and tumor size groups in nasopharyngeal carcinoma. The expressions of Twist and HIF-1α mRNA and protein in different lymph node metastasis, clinical stage and pathological grading group The difference was statistically significant. There was a positive correlation between Twist mRNA and protein expression, mRNA and protein expression of HIF-1α, Tw ist and protein expression of HIF-1α, Tw ist and HIF-1α mRNA in NPC tissues. Conclusions Twist and HIF-1α are highly expressed in NPC tissues, and they are correlated with each other. HIF-1α protein may be one of the ways to promote the invasion and metastasis of nasopharyngeal carcinoma by up-regulating Twist expression.
