目的:观察芪仙汤对哮喘小鼠肺组织中FIZZ1基因及蛋白表达水平的影响,探讨芪仙汤治疗哮喘的作用机制。方法:选用30只健康雌性BALB/c小鼠,采用随机数字表分为正常对照组(对照组)、支气管哮喘模型组(模型组)、芪仙汤治疗组(治疗组)。除对照组外均采用卵白蛋白(OVA)建立支气管哮喘小鼠模型。各组分别相应干预灌胃2周。从小鼠的一般情况、肺组织病理形态学(HE染色病理切片)指标评价支气管哮喘小鼠模型;采用RT-PCR检测肺组织FIZZ1基因mRNA表达水平,Western blot和免疫组化法检测肺组织FIZZ1蛋白表达水平。结果:HE染色结果显示,模型组小鼠肺组织较正常组嗜酸性粒细胞等炎症细胞浸润明显,气道壁及支气管平滑肌增厚明显;治疗组哮喘病理改变明显改善。RT-PCR、Western blotting、免疫组化结果均显示模型组FIZZ1表达水平显著高于对照组,治疗组小鼠肺组织中FIZZ1表达水平明显低于模型组,差异均有统计学意义(P<0.05,P<0.01)。结论:芪仙汤可能是通过下调小鼠肺组织FIZZ1表达起到治疗支气管哮喘的作用。
Objective: To observe the effect of Qixian decoction on the expression of FIZZ1 gene and protein in lung tissue of asthmatic mice, and explore the mechanism of Qixian Decoction in treating asthma. Methods: Thirty healthy female BALB / c mice were randomly divided into normal control group (control group), bronchial asthma model group (model group) and Qixian Decoction group (treatment group). In addition to the control group were used ovalbumin (OVA) to establish a mouse model of bronchial asthma. Each group corresponding to the corresponding gavage for 2 weeks. The expression of FIZZ1 mRNA in lung tissue was detected by RT-PCR and the expression of FIZZ1 protein in lung tissue was detected by Western blot and immunohistochemistry The expression level. Results: The results of HE staining showed that inflammatory cells such as eosinophils infiltrated obviously in the lung tissue of model mice, and the thickening of airway wall and bronchial smooth muscle was obvious. The pathological changes of asthma in the treatment group were significantly improved. The results of RT-PCR, Western blotting and immunohistochemistry showed that the expression of FIZZ1 in the model group was significantly higher than that in the control group. The expression of FIZZ1 in the lung tissue of the treated group was significantly lower than that of the model group (P <0.05) , P <0.01). Conclusion: Qixian Decoction may play an important role in the treatment of bronchial asthma by down-regulating the expression of FIZZ1 in lung tissue of mice.
