目的研究HIP-55在β-肾上腺素受体激动剂异丙基肾上腺素(Isoproterenol hydrochloride,ISO)诱导心脏成纤维细胞活性氧(reactive oxygen species,ROS)产生中的作用。方法分离培养乳大鼠心脏成纤维细胞,利用HIP-55腺病毒和HIP-55-shRNA腺病毒感染乳大鼠心脏成纤维细胞,获得过表达HIP-55或敲减HIP-55的心脏成纤维细胞。ISO刺激后,采用红色荧光探针dihydroethidium(DHE)以及绿色荧光探针二氯荧光黄双乙酸钠(2’,7’-dichlorofluoresce in diacetate,DCFH-DA)的方法检测细胞ROS的产生量。结果过表达HIP-55明显的抑制心脏成纤维细胞ROS的产生,相反敲减HIP-55明显促进心脏成纤维细胞ROS的产生。结论 HIP-55明显抑制ISO诱导的乳大鼠成纤维细胞的ROS的产生。
Objective To investigate the role of HIP-55 in the production of reactive oxygen species (ROS) in cardiac fibroblasts induced by β-adrenergic receptor agonist isoproterenol hydrochloride (ISO). Methods Fibroblasts were isolated from cultured rat cardiac fibroblasts and infected with HIP-55 adenovirus and HIP-55-shRNA adenovirus to induce cardiac fibroblasts. HIF-55 overexpression or HIP-55 knockdown was performed on cardiac fibroblasts cell. After ISO stimulation, ROS production was measured by using dihydroethidium (DHE) as a green fluorescent probe and DCFH-DA as a green fluorescent probe. Results Overexpression of HIP-55 significantly inhibited ROS production in cardiac fibroblasts. In contrast, knockdown of HIP-55 markedly promoted ROS production in cardiac fibroblasts. Conclusion HIP-55 significantly inhibits ISO-induced ROS production in rat fibroblasts.
