目的:探讨六味地黄汤治疗Ig A肾病的作用机制。方法:实验动物分为正常组、模型组、雷公藤组和六味地黄汤组。除正常组外,均采用牛血清白蛋白(BSA)+脂多糖(LPS)+四氯化碳(CCl4)方法建立实验性Ig AN模型。造模成功后连续予以相应药物灌胃4周,正常组予等体积0.9%Na Cl溶液灌胃。测定各组大鼠24 h尿蛋白;检测血肌酐及尿素氮的含量;用HE和Masson染色法观察肾脏组织形态学改变;免疫组织化学方法检测大鼠肾组织中IL-6和TNF-α的表达。结果:与正常组比较,模型组的24 h尿蛋白明显升高、IL-6和TNF-α表达明显升高(P<0.05);与模型组比较,雷根藤组和六味地黄汤组24 h尿蛋白明显降低,IL-6和TNF-α表达明显降低,差异均有统计学意义(P<0.05);各组大鼠肾功能结果差异无统计学意义(P>0.05);雷公藤组及六味地黄汤组肾小球系膜细胞增生和系膜基质增多情况较模型组明显减轻。结论:六味地黄汤抑制IL-6和TNF-α的表达可能是其治疗Ig A肾病的作用机制之一。
Objective: To explore the mechanism of Liuwei Dihuang Decoction in treating IgA nephropathy. Methods: Experimental animals were divided into normal group, model group, Tripterygium wilfordiii group and Liuweidihuang decoction group. In addition to the normal group, experimental Ig AN model was established by using BSA + LPS + CCl4. Successfully made after the corresponding drug gavage for 4 weeks, the normal group was given an equal volume of 0.9% Na Cl solution gavage. The 24 h urinary protein of rats was measured. The levels of serum creatinine and urea nitrogen were measured. The morphological changes of kidney were observed by HE and Masson staining. The levels of IL-6 and TNF-α in renal tissue were detected by immunohistochemistry expression. Results: Compared with the normal group, 24 h urinary protein in the model group increased significantly and the expression of IL-6 and TNF-α significantly increased (P <0.05). Compared with the model group, (P <0.05). There was no significant difference in renal function between the two groups (P> 0.05). The contents of IL-6 and TNF- And Liu Wei Di Huang Tang group glomerular mesangial cell proliferation and mesangial matrix increased significantly less than the model group. Conclusion: The inhibitory effect of Liuwei Dihuang decoction on the expression of IL-6 and TNF-α may be one of its mechanisms in the treatment of IgA nephropathy.
