目的观察小鼠脑缺血再灌注以及高压氧治疗后海马脑红蛋白表达的变化,探讨高压氧对脑红蛋白表达的影响及其意义。方法雌性C57BL/6N小鼠随机分为假手术对照组、缺血再灌注48h组(48h I/R组)及缺血再灌注48h加高压氧治疗组(48h HBO组)。对各组小鼠脑组织神经元进行HE及免疫组织化学染色、图像学分析及统计学处理。结果48h I/R组及48h HBO组的脑红蛋白表达均高于假手术组(P<0.01和P<0.001),48hHBO组吸光度高于48h I/R组(P<0.01)。结论高压氧对缺血性脑损伤有显著治疗作用,提高脑红蛋白的表达水平可能是其治疗的机制之一。
Objective To observe the changes of hippocampal neuroglobin after cerebral ischemia-reperfusion and hyperbaric oxygen therapy in mice and to explore the effect of hyperbaric oxygen on the expression of neuroglobin and its significance. Methods Female C57BL / 6N mice were randomly divided into sham operation control group, ischemia / reperfusion 48h group (48h I / R group) and ischemia / reperfusion 48h hyperbaric oxygen therapy group (48h HBO group). The neurons in each group of mice were subjected to HE and immunohistochemical staining, image analysis and statistical analysis. Results The expression of neuroglobin in 48h I / R group and 48h HBO group was significantly higher than that in sham operation group (P <0.01 and P <0.001). The absorbance in 48h HBO group was higher than that in 48h I / R group (P <0.01). Conclusion Hyperbaric oxygen has a significant therapeutic effect on ischemic brain injury. Increasing the expression of neuroglobin may be one of the mechanisms of its treatment.
