目的:观察四君子汤对脾虚证大鼠血清生长激素释放肽(Ghrelin)、胃窦肥大细胞(MC)数量及其脱颗粒的影响,探讨四君子汤健脾益气的作用机制。方法:将40只SD大鼠随机分为4组:正常组、模型组、中药组、西药组,利用耗气破气加饥饱失常法加劳倦法塑造脾虚证大鼠模型14d。造模成功后中药组予四君子汤灌胃,西药组予莫沙必利灌胃,模型组予0.9%氯化钠溶液灌胃,连续15d。采用双抗体两步夹心酶联免疫吸附法(ELISA)检测血清Ghrelin的含量,光镜高倍镜(40×10)下观察随机选5个连续视野,进行MC计数及MC脱颗粒计数。结果:与正常组相比,模型组大鼠血清Ghrelin含量明显降低(P<0.05)、胃窦黏膜MC数量及其脱颗粒数量显著升高(P<0.05);与模型组比较,中药组与西药组血清Ghrelin含量均有显著的升高,胃窦MC数量及其脱颗粒数量显著下降(P<0.05)。结论:四君子汤可以明显改善脾虚证大鼠症状,其可能是通过提高血清Ghrelin含量及减少MC数量及其脱颗粒来发挥机制。
Objective: To observe the effect of Sijunzi Decoction on serum ghrelin (Ghrelin), mast cells (MC) and degranulation of gastric antrum in rats with spleen deficiency syndrome and to explore the mechanism of Sijunzi Decoction for invigorating spleen and qi. Methods: Forty Sprague-Dawley rats were randomly divided into 4 groups: normal group, model group, TCM group and western medicine group. The model group was given gavage with 0.9% sodium chloride solution for 15 days. Serum levels of Ghrelin were detected by double-antibody sandwich enzyme-linked immunosorbent assay (ELISA). Five consecutive fields of vision were selected randomly under light microscope (40 × 10) to count MC count and MC degranulation. Results: Compared with the normal group, the Ghrelin level in the model group was significantly decreased (P <0.05), and the number of MC in the gastric mucosa and the number of degranulation were significantly increased (P <0.05). Compared with the model group, Western medicine group serum Ghrelin levels were significantly increased, the number of gastric antrum and the number of degranulation significantly decreased (P <0.05). Conclusion: Sijunzi Decoction can significantly ameliorate the symptoms of rats with spleen deficiency syndrome. It may play a role in improving the serum Ghrelin content and reducing the number of MC and its degranulation.
