目的了解浙江省不同地区淡色库蚊钠离子通道(vssc)基因多态性分布特征。方法采集杭州、舟山、湖州、金华等市区淡色库蚊样本,PCR扩增vssc基因片段,扩增产物进行基因单链构象多态性(single strand conformation polymorphism,SSCP)分析。对不同的SSCP型进行测序,并进行氨基酸序列预测。结果四地市淡色库蚊现场品系以及敏感品系vssc基因片段经SSCP分析共发现7个SSCP型,分别属于3个不同的氨基酸序列型,即野生型、L1014S突变型以及L1014F突变型。结论浙江省淡色库蚊vssc基因存在较高的多态性,使其在面对菊酯类等相关杀虫剂选择压力时具有较高的耐受度,进而可能会增大相关杀虫抗性防制的难度。
Objective To investigate the distribution of polymorphism of vssc gene in Culex pipiens pallens in different areas of Zhejiang Province. Methods The specimens of Culex pipiens pallens collected from Hangzhou, Zhoushan, Huzhou and Jinhua were amplified by polymerase chain reaction (PCR). The single strand conformation polymorphism (SSCP) analysis of the amplified products was carried out. Different SSCP types were sequenced and amino acid sequence prediction was performed. Results Seven SSCP patterns were found in the field strains of Culex pipiens pallens and the susceptible strain vssc gene fragments in the four cities, belonging to three different amino acid sequences, namely wild type, L1014S mutant and L1014F mutant. Conclusion The vssc gene of Culex pipiens pallens in Zhejiang Province has a high polymorphism, which makes it more tolerant to the selection pressure of insecticides such as pyrethrum and may increase the insecticidal resistance Prevention of the difficulty.
