目的探讨组织蛋白酶D(Cathepsin D,CD)和第10号染色体磷酸酶和张力蛋白同源丢失性基因(phosphatase and tension homolog deleted on chromosome10,PTEN)表达与人胶质瘤恶性程度和侵袭性之间的关系。方法采用免疫组织化学法和Western blot法检测46例胶质瘤标本和10例正常脑组织标本中CD及PTEN的表达情况。结果 CD阳性染色主要定位于细胞质内,呈棕黄色细颗粒状,在高分化组(Ⅰ-Ⅱ级)中呈散在的弱阳性表达,表达率为50%(14/28);而在低分化组(Ⅲ-Ⅳ级),表达明显增强,表达率为83.3%(15/18),与高分化组有显著差异(P<0.05)。PTEN阳性染色主要定位于细胞质内,46例胶质瘤中PTEN阳性表达18例(39.1%),高分化组(Ⅰ-Ⅱ级)中15/28例(53.6%)明显高于低分化组(Ⅲ-Ⅳ级)3/18例(16.7%,P<0.05)。结论 CD和PTEN的表达对分析、判断胶质瘤分级及恶性转化具有一定意义。
Objective To investigate the relationship between the expression of Cathepsin D (CD), phosphatase and tension homolog deleted on chromosome 10 (PTEN) and the malignancy and invasiveness of human glioma Relationship. Methods Immunohistochemistry and Western blot were used to detect the expression of CD and PTEN in 46 glioma specimens and 10 normal brain tissues. Results CD positive staining mainly localized in the cytoplasm, showing a brownish-yellow fine granular, weakly positive expression in the well-differentiated group (grade Ⅰ-Ⅱ), the expression rate was 50% (14/28) Group (grade Ⅲ-Ⅳ), the expression was significantly enhanced, the expression rate was 83.3% (15/18), significantly different from the well-differentiated group (P <0.05). Positive staining of PTEN was mainly located in the cytoplasm. Positive expression of PTEN in 46 gliomas was found in 18 cases (39.1%), while in 15 cases (53.6%) in well-differentiated group (Ⅰ-Ⅱ) was significantly higher than that in poorly differentiated group Ⅲ-Ⅳ grade) 3/18 cases (16.7%, P <0.05). Conclusion The expression of CD and PTEN has some significance for the analysis and judgment of glioma grade and malignant transformation.
