目的:观察养心通脉颗粒Ⅱ方对气虚血瘀型心衰大鼠的作用机理。方法:通过多因素诱导复制中医气虚血瘀型大鼠模型,利用阿霉素诱导复制心衰大鼠模型。60只SD雌性大鼠随机分为养心通脉颗粒Ⅱ方组、依那普利组、模型组及空白组各15只。造模后养心通脉颗粒Ⅱ方组、依那普利组分别予养心通脉颗粒Ⅱ方、依那普利灌胃治疗,模型组、空白对照组予生理盐水灌胃治疗,均灌胃4周。末次给药后24h采集标本,用酶联免疫法测定各组大鼠血管紧张素Ⅱ(AngⅡ)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的浓度;处死大鼠,取大鼠心脏,测大鼠左室重量指数,用tunel法检测心肌细胞凋亡指数。结果:养心通脉颗粒Ⅱ方及依那普利组与模型组比较,均可有效降低心衰大鼠血清AngⅡ、TNF-α、IL-6水平(P<0.05);养心通脉颗粒Ⅱ方组及依那普利组与模型组比较,左室重量指数、心肌细胞凋亡指数明显降低(P<0.05)。结论:养心通脉颗粒Ⅱ方对气虚血瘀型大鼠的治疗作用,可能与其降低大鼠血清中神经内分泌细胞因子AngⅡ、TNF-α、IL-6的水平,降低左室重量指数,抑制心肌细胞凋亡有关。
Objective: To observe the mechanism of Yangxin Tongmai Granule (Ⅱ) on heart failure rats with qi deficiency and blood stasis syndrome. Methods: To induce the rat model of Qi-deficiency and blood-stasis syndrome induced by multiple factors and to use doxorubicin to induce the rat model of heart failure. Sixty SD female rats were randomly divided into Yangxintongmai granule Ⅱ group, enalapril group, model group and blank group 15 each. After modeling, Yangxintongmai granuleⅡgroup and enalapril group were treated with Yangxintongmai granuleⅡand enalapril intragastrically, respectively. The model group and blank control group were treated with saline, Stomach for 4 weeks. The samples were collected 24 h after the last administration and the concentrations of angiotensin Ⅱ, tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were measured by enzyme-linked immunosorbent assay The rats were sacrificed, the heart of the rats was taken, the left ventricular mass index was measured, and the cardiomyocyte apoptosis index was detected by tunel method. Results: Compared with model group, Yangxintongmai Granules Ⅱ and Enalapril groups could effectively reduce the levels of serum AngⅡ, TNF-α and IL-6 in heart failure rats (P <0.05) Compared with the model group, the left ventricular mass index and cardiomyocyte apoptotic index in Group Ⅱ and Enalapril were significantly decreased (P <0.05). Conclusion: The therapeutic effect of Yangxintongmai Granule Ⅱ on Qi-deficiency and Blood-stasis rats may be related to the decrease of serum levels of neuroendocrine cytokines AngⅡ, TNF-α and IL-6, the decrease of left ventricular mass index, Cardiomyocyte apoptosis related.
