目的研究铜绿假单胞菌在霍乱弧菌分离、培养过程中对霍乱弧菌的生长是否存在拮抗作用。方法挑取典型的霍乱弧菌和铜绿假单胞菌共同增菌、分离培养,观察分离、培养的结果,并分别用MH琼脂和碱性琼脂进行药敏试验。结果 10株霍乱弧菌和10株铜绿假单胞菌对应等量混合接种于碱性蛋白胨水,37℃培养6h后划线接种碱性琼脂平板,只检出1例霍乱弧菌(O139),碱性蛋白水37℃培养18h后划线接种碱性琼脂平板则未检出霍乱弧菌;将10株霍乱弧菌接种于不同稀释度灭菌前和灭菌后铜绿假单胞菌菌液中,37℃培养6h后划线接种碱性琼脂平板,只有当灭菌前菌液稀释度(1∶12、灭菌后菌液稀释度(1∶4时才能检出霍乱弧菌;强力霉毒和四环素在M-H琼脂和碱性琼脂上对霍乱弧菌抑菌圈的直径分别为16.5mm、11.5mm和7.0mm、6.5mm。结论铜绿假单胞菌及其代谢产物对霍乱弧菌的生长存在拮抗作用;在碱性环境中,强力霉素和四环素对霍乱弧菌的抑制作用明显不如在中性环境中的抑制作用。
Objective To study whether Pseudomonas aeruginosa could antagonize the growth of Vibrio cholerae during the isolation and culture of Vibrio cholerae. Methods Typical strains of Vibrio cholera and Pseudomonas aeruginosa were selected and cultured. Isolation and culture were observed. The results of isolation and culture were observed. Susceptibility test was performed on MH agar and alkaline agar respectively. Results 10 strains of Vibrio cholerae and 10 strains of Pseudomonas aeruginosa were inoculated into alkaline peptone water in the same amount. After incubation at 37 ℃ for 6h, alkaline agar plates were streaked and only one case of Vibrio cholerae (O139) Vibrio cholerae was not detected after alkaline protein water was incubated at 37 ℃ for 18 hours and then inoculated with alkaline agar plate. Vibrio cholerae were inoculated in different dilutions of Pseudomonas aeruginosa before and after sterilization , And incubated at 37 ℃ for 6h, then inoculated with alkaline agar plate, only when the dilution of bacteria before sterilization (1:12, after dilution of bacteria diluted (1: 4 to detect Vibrio cholerae; And tetracycline against Vibrio cholerae on MH agar and alkaline agar were 16.5mm, 11.5mm and 7.0mm, 6.5mm, respectively.Conclusion Pseudomonas aeruginosa and its metabolites have a positive effect on the growth of V. cholerae Antagonistic effect. In alkaline environment, the inhibitory effect of doxycycline and tetracycline on Vibrio cholerae is obviously not as good as that in neutral environment.
