目的利用C6胶质瘤干细胞建立wistar大鼠脑胶质瘤模型,观察肿瘤生长规律及病理特征,探讨利用肿瘤干细胞构建模型的优势。方法体外提取并培养C6细胞系中胶质瘤干细胞,应用立体定向法在鼠脑右侧尾状核区接种104个胶质瘤干细胞,术后连续观察大鼠生存状态和生存时间,分时段行鼠脑MRI检查、病理HE切片及GFAP免疫组织化学检查,观察肿瘤生长及病理特征。结果利用本方法建立模型成瘤率为100%,术后大鼠恢复快,术后反应轻,未见颅外转移,生存期稳定,重复性好,肿瘤生长规律、影像学特征及组织病理特点与人脑胶质瘤相似。结论利用胶质瘤干细胞建立的大鼠脑胶质瘤模型稳定可靠,肿瘤生长更贴近胶质瘤在脑内自然发生的特点,且较传统方法脑内注射体积小,模型制作时间短,术后恢复快,成瘤率更高等优点,为研究胶质瘤干细胞在肿瘤形成过程中的生长方式、分子机制及实验性治疗提供了一个较为理想的模型。
OBJECTIVE: To establish a glioma model of wistar rat by using C6 glioma stem cells. Observe the growth and pathological features of the tumor and explore the advantages of using the model of cancer stem cells. Methods Glioma stem cells were isolated and cultured in vitro. 104 glioma stem cells were inoculated to the right caudate nucleus caudatum by stereotactic method. The survival state and survival time of rats were observed continuously after operation. Brain MRI, pathological HE sections and GFAP immunohistochemistry to observe the tumor growth and pathological features. Results The model established by this method had a tumorigenic rate of 100%. The postoperative recovery of the rats was rapid, the postoperative reaction was mild, there was no extracranial metastasis, the survival period was stable, the reproducibility was good, the tumor growth pattern, the imaging features and the histopathological features Similar to human glioma. Conclusion The model of rat glioma established by glioma stem cells is stable and reliable. The tumor growth is more close to the nature of glioma occurring in the brain. Compared with the traditional method, the brain injection volume is small, the model making time is short, Faster recovery, higher tumor formation rate, etc. It provides an ideal model for studying the growth pattern, molecular mechanism and experimental treatment of glioma stem cells in the process of tumor formation.
