目的探究mCD99L2基因沉默对小鼠B淋巴瘤细胞系A20细胞生物学特性的影响。方法采用MTT法和流式细胞仪检测mCD99L2基因沉默前后干扰组A20-LV-mCD99L2和未干扰组A20细胞的增殖率和细胞周期;Transwell法测定两组细胞的体外侵袭能力;采用裸鼠与BALB/c鼠皮下成瘤实验观察比较两组细胞在体内的成瘤时间和成瘤率。结果MTT法显示干扰组A20-LV-mCD99L2增殖率为(0.61±0.12),低于未干扰组增殖率(0.75±0.20),差异有统计学意义(P=0.000);干扰组A20-LV-mCD99L2处于G2期的细胞比例为(10.58±4.97),高于未干扰组(3.33±1.31),差异有统计学意义(P=0.009);Transwell法显示干扰组A20-LV-mCD99L2运动能力较未干扰组下降。裸鼠皮下成瘤时间干扰组(13.33±4.63)d长于未干扰组(9.50±2.90)d,成瘤率均为100%;BALB/c鼠皮下成瘤时间干扰组(10d)长于未干扰组(7.0±0.82)d,成瘤率干扰组为14.3%,显著低于未干扰组(100%),差异有统计学意义(P=0.000)。结论mCD99L2基因沉默可抑制小鼠B淋巴瘤细胞系A20细胞的生物学行为,降低其在体外的增殖能力和运动能力,显著降低其在BALB/c鼠体内的成瘤率。
Objective To investigate the effect of mCD99L2 gene silencing on the biological characteristics of mouse B lymphoma cell line A20. Methods MTT assay and flow cytometry were used to detect the proliferation and cell cycle of A20-LV-mCD99L2 and untransfected A20 cells before and after silencing mCD99L2 gene silencing. Transwell assay was used to determine the in vitro invasiveness of the two cell lines. / c mouse into the tumor under the experimental observation The two groups of cells in vivo tumor formation time and tumor formation rate. Transwell assay showed that the ability of A20-LV-mCD99L2 Interference group decreased. (13.33 ± 4.63) d in nude mice was longer than that in non-interfering group (9.50 ± 2.90) d, and the tumorigenic rate was 100%. The subcutaneous tumorigenic time in BALB / c mice was longer than that in non-interfering group (7.0 ± 0.82) d. The rate of tumorigenesis in intervention group was 14.3%, which was significantly lower than that in non-interference group (100%). The difference was statistically significant (P = 0.000). Conclusion Silencing of mCD99L2 can inhibit the biological behavior of mouse B lymphoma cell line A20, reduce its proliferation and exercise capacity in vitro, and significantly reduce its tumorigenic rate in BALB / c mice.
