目的:体外探讨肿瘤坏死因子-α(TN F-α)和白细胞介素-6(IL-6)对关节软骨细胞胶原表型表达的影响。方法:分离成人正常关节软骨细胞,用 TN F-α单独或与IL-6联合干预培养,采用形态学观察、流式细胞仪分析、Nort hern杂交技术,检测关节软骨细胞表型。结果:TNF-α可促使软骨细胞向成纤维细胞样细胞形态转化及分裂、增殖,抑制其表达特异性Ⅱ型胶原,而诱导其表达非特异性Ⅲ型胶原;IL-6有协同 TN F-α干扰软骨细胞表型表达的作用。结论: TNF-α和IL-6是参与关节软骨细胞变性的介导因素。
Objective: To investigate the effect of tumor necrosis factor-α (TN F-α) and interleukin-6 (IL-6) on the expression of collagen phenotype in articular chondrocytes in vitro. METHODS: Adult normal articular chondrocytes were isolated and cultured with either TN F-alpha alone or in combination with IL-6. Morphological observation, flow cytometric analysis, and North hern hybridization were performed to detect the phenotype of articular chondrocytes. RESULTS: TNF-α promoted the transformation and proliferation of chondrocytes into fibroblast-like cells, inhibited their expression of type II collagen, and induced the expression of non-specific type III collagen; IL-6 had synergistic TN F-α Interferes with the phenotypic expression of chondrocytes. Conclusion: TNF-α and IL-6 are involved in the degeneration of articular cartilage cells.
