角朊细胞激活对T淋巴细胞增殖和细胞因子分泌的作用。TPA和IFN γ联合作用 ,激活角朊细胞表达表面分子 ,FACS检测MHCI、II类分子和CD80、CD86分子水平 ,RT PCR检测B7 H1共刺激分子表达。由激活角朊细胞提供T淋巴细胞激活的第二信号 ;绵羊红细胞花环沉淀分离纯化T细胞 ,抗CD3单抗提供第一信号 ,建立体外混合培养系统 ,3 H TdR渗入法检测T细胞增殖 ,ELISA检测培养上清细胞因子浓度。结果显示 ,TPA和IFN γ联合作用可诱导角朊细胞表达B7 H1共刺激分子 ,并上调其MHCII类分子。以B7 H1为第二信号 ,可以协同抗CD3单抗对T细胞的增殖作用 ,并呈现独特的细胞因子分泌格局。因此角朊细胞激活后表达B7 H1共刺激分子 ,此第二信号可刺激T淋巴细胞增殖 ,对其功能分化具有调节作用
Effect of Keratinocyte Activation on T Lymphocyte Proliferation and Cytokine Secretion. The combination of TPA and IFNγ activated keratinocytes to express surface molecules. The levels of MHC class II molecules, CD80 and CD86 were detected by FACS. The expression of costimulatory molecules B7 H1 was detected by RT PCR. Activated keratinocytes provide the second signal of T lymphocyte activation; sheep erythrocyte wreath precipitation separation and purification of T cells, anti-CD3 monoclonal antibody to provide the first signal to establish an in vitro mixed culture system, 3 H TdR infiltration assay T cell proliferation, ELISA Detection of culture supernatant cytokine concentration. The results showed that the combination of TPA and IFN γ induced keratinocytes to express B7 H1 co-stimulatory molecules and up-regulated their MHC class II molecules. Taking B7 H1 as the second signal, synergistic anti-CD3 monoclonal antibody can proliferate T cells and exhibit unique cytokine secretion pattern. Therefore keratinocytes express B7 H1 co-stimulatory molecules after activation, and this second signal can stimulate the proliferation of T lymphocytes and regulate the differentiation of keratinocytes
