我们已经报告了钙拮抗剂维拉帕米(Verapamil,Ver)、硝苯啶(nifedipir.Nif)及尼卡地平(nicardipine,Nic)具有抗炎作用,并且降低炎症组织里炎症介质PGE_2含量。目前认为在炎症、缺血再灌注、休克等多种病理过程中,氧自由基对组织细胞的损害起重要作用。过氧化氢(hydrogen peroxide,H_2O_2)是一种重要的氧自由基,为了探讨钙拮抗剂类药物的抗炎机理,本研究在离体实验条件下,观察和比较了4种二氢吡啶类钙拮抗剂Nic、Nif、尼莫地平(niroodipine,Nim)、尼群地平(ni(?)endipine,Nit)、以胶Ver对小鼠腹腔巨噬细胞释放H_2O_2的影响。 H_2O_2采用荧光测定法。其原理是在辣根过氧化酶的存在下,H_2O_2氧化莨菪亭,使后者失去荧光,通过介质甲莨菪亭荧光强度的衰减,可定量测出H_2O_2的释放量。来自C57BL/6小鼠的腹腔巨噬细胞和不同浓度的钙拮抗剂在37℃温育3h后,巨
