目的:探讨Notch信号分子及下游靶基因在人NK/T细胞淋巴瘤、NK细胞淋巴瘤、T细胞淋巴瘤和B细胞淋巴瘤细胞中的表达,阐明Notch信号分子在NK细胞肿瘤中的作用及意义。方法:选择NK/T细胞淋巴瘤细胞SNK-6、NK细胞淋巴瘤细胞YTS、人急性T淋巴细胞白血病细胞Jurkat和人Burkitt’s淋巴瘤细胞Raji,通过实时荧光定量PCR技术检测上述细胞中Notch信号分子的表达水平。结果:Notch 1,3,4信号分子在YTS和SNK-6细胞中表达量均不同程度高于Raji和Jurkat淋巴瘤细胞(P<0.01),但是在SNK-6中的表达量均不同程度低于在YTS细胞中的表达(P<0.01)。Notch 2信号分子在YTS、SNK-6细胞中的表达量高于Jurkat细胞(P<0.01),但是与Raji细胞比较差异无统计学意义(P>0.05)。Notch下游靶基因Hes1的表达量,YTS细胞高于Raji和Jurkat细胞(P<0.05,P<0.01),而SNK-6细胞则低于Raji和Jurkat细胞(P<0.01)。结论:在NK细胞肿瘤细胞中Notch信号分子表达异常,且明显区别于B、T细胞淋巴瘤细胞。
Objective: To investigate the expression of Notch signaling molecule and its downstream target genes in human NK / T cell lymphoma, NK cell lymphoma, T-cell lymphoma and B-cell lymphoma and to elucidate the role of Notch signaling in NK cell tumor and significance. Methods: SNK-6 of NK / T cell lymphoma, YTS of NK cell lymphoma, Jurkat of human acute lymphoblastic leukemia, and Raji of human Burkitt’s lymphoma were selected to detect Notch signaling molecules in these cells by real-time fluorescence quantitative PCR The level of expression. Results: The expression of Notch 1,3,4 signaling molecules in both YTS and SNK-6 cells was significantly higher than that in Raji and Jurkat lymphoma cells (P <0.01), but not in SNK-6 In YTS cells (P <0.01). The expression of Notch 2 in YTS and SNK-6 cells was higher than that in Jurkat cells (P <0.01), but there was no significant difference compared with Raji cells (P> 0.05). The expression of Notch downstream target gene Hes1 was higher in YTS cells than in Raji and Jurkat cells (P <0.05, P <0.01), but lower in SNK-6 cells than in Raji and Jurkat cells (P <0.01). CONCLUSIONS: Notch signaling molecules are abnormally expressed in NK cell tumor cells and are significantly different from B and T cell lymphoma cells.
