目的:探讨炎症状态和妊娠期高血压疾病(HDCP)发生的关系,初步筛选HDCP早期标志物。方法:采用病例对照的方法研究601例HDCP患者和1 053例正常妊娠妇女血液系统白细胞参数,分析其在两组中的变化趋势;采用蛋白芯片筛选507种蛋白在1∶1匹配的8例妊娠20周前孕妇血浆中的表达情况。结果:妊娠20周前,HDCP患者的白细胞参数比对照组明显升高,两组中有5类31种蛋白表达水平差异有统计学意义:除GH升高外,血管生长因子(血管生成素样蛋白2、EG-VEGF和PIGF)、炎症反应因子(IL-1 sRⅡ、TWEAK、IFN-α/β、IL-2、IL-6 R、MIP-3β、RANK、CCR6、TLR4、OX40配体、趋化因子、白细胞衍生趋化因子2、TNF-α)、生长因子(IGF-ⅡR、IGFBP-1、FGF-21、血小板反应蛋白-4、GM-CSF)、基质金属蛋白(MMP-11、MMP-1、MMP-10)和其他蛋白(食欲素B、TMEFF2、卷曲蛋白-6、XEDAR、抑制素-A和泛激素)表达降低。结论:证实炎症与HDCP发生有关,筛选出的31种蛋白可能是HDCP潜在的早期标志物。
Objective: To investigate the relationship between inflammatory status and occurrence of HDCP in pregnancy, and to screen the early HDCP markers. Methods: The blood leukocyte parameters of 601 HDCP patients and 1053 normal pregnant women were studied by case-control method, and the trend of leukocyte count in both groups were analyzed. Protein chips were used to screen 507 proteins in 1: 1 matched 8 cases of pregnancy The expression of maternal plasma 20 weeks ago. Results: Before pregnancy, the white blood cell parameters of HDCP patients were significantly higher than that of the control group. There were 5 types of 31 proteins in the two groups. There was significant difference between the two groups (P <0.05). Except GH, the levels of angiogenin Protein 2, EG-VEGF and PIGF), inflammatory response factors (IL-1 sRII, TWEAK, IFN- alpha / beta, IL- 2, IL- 6R, MIP-3beta, RANK, CCR6, TLR4, OX40 ligand, Chemokine 2, TNF-α), growth factor (IGF-ⅡR, IGFBP-1, FGF-21 and thrombospondin-4, GM-CSF), matrix metalloproteinases MMP-1, MMP-10) and other proteins (orexin B, TMEFF2, Frizzled-6, XEDAR, inhibin-A and pan-hormones). Conclusion: It is confirmed that inflammation is related to the occurrence of HDCP. The 31 proteins screened out may be potential early markers of HDCP.
