目的了解在腹腔注射家蝇幼虫分泌物抗菌肽的干预作用下,大肠埃希菌感染小鼠的外周血细胞过氧化物酶及多项血液指标的变化。方法将实验小鼠随机分为空白对照、模型对照、抗菌肽干预3个组,并分别于处理后24 h、48 h、72 h取样分析;外周血细胞过氧化物酶染色采用四甲基联苯胺法,全自动血细胞分析仪检测血红蛋白、血小板、白细胞、红细胞计数及白细胞细胞分类情况。结果在各检测指标中,白细胞数、中性粒细胞绝对值在实验处理间具有显著性差异;感染72 h抗菌肽干预组白细胞数高于模型对照组(P<0.05);与空白对照组比较,抗菌肽干预组和模型对照组各检测时间点白细胞数均有显著性差异(P<0.05);感染72 h抗菌肽干预组中性粒细胞绝对值显著高于模型对照组(P<0.05);血细胞过氧化物酶染色及其余指标在处理因素间无统计学差异。结论腹腔注射家蝇幼虫分泌物抗菌肽对小鼠大肠埃希菌感染具有一定的干预作用。
Objective To investigate the changes of peripheral blood peroxidase and blood parameters in Escherichia coli-infected mice under the action of intraperitoneal injection of antibacterial peptides from housefly larvae secretions. Methods The experimental mice were randomly divided into three groups: blank control, model control and antibacterial peptide intervention, and were sampled at 24, 48 and 72 h after treatment respectively. Peripheral blood cell peroxidase staining was performed with tetramethylbenzidine Method, automatic hemoglobin analyzer hemoglobin, platelets, white blood cells, red blood cell count and white blood cell classification. Results The absolute numbers of leukocytes and neutrophils were significantly different between treatments in each test index. The number of leukocytes in the antimicrobial peptide intervention group at 72 h after infection was higher than that in the model control group (P <0.05). Compared with the blank control group (P <0.05). The absolute value of neutrophils in the antimicrobial peptide intervention group at 72 h was significantly higher than that in the model control group (P <0.05), and the number of white blood cells in the antimicrobial peptide intervention group and the model control group at each detection time point was significantly different ; Peroxidase staining of blood and the remaining indicators in the treatment of factors between no significant difference. Conclusion Intraperitoneal injection of housefly larvae secreted antimicrobial peptides have some intervention effects on E. coli infection in mice.
