目的:考察黄连素是否通过调节GRP78表达而促进SW480细胞凋亡。方法:MTT法检测不同浓度黄连素对结肠癌细胞增殖的影响,流式细胞术检测细胞凋亡、Western blot法检测细胞GRP78、Bcl-2、Bax、GRP94的表达。结果:与对照组比较,不同浓度黄连素可呈浓度依赖性抑制SW480和HT-29细胞的增殖,其对SW480细胞的作用较强。黄连素可促进SW480细胞凋亡,抑制其GRP78表达,使Bax表达上调,Bcl-2表达下调。Bi X激活GRP78表达后,受黄连素作用的SW480细胞凋亡率降低,Bax表达下调,Bcl-2表达上调,但不同处理组GRP94表达均无显著变化。结论:黄连素可通过下调GRP78的表达来促进SW480细胞凋亡。
Objective: To investigate whether berberine can promote the apoptosis of SW480 cells by regulating the expression of GRP78. Methods: The effects of different concentrations of berberine on the proliferation of colon cancer cells were detected by MTT assay. The apoptosis of colon cancer cells was detected by flow cytometry. The expressions of GRP78, Bcl-2, Bax and GRP94 were detected by Western blot. Results: Compared with the control group, different concentrations of berberine could inhibit the proliferation of SW480 and HT-29 cells in a concentration-dependent manner, which had a strong effect on SW480 cells. Berberine can promote the apoptosis of SW480 cells, inhibit the expression of GRP78, up-regulate the expression of Bax and down-regulate the expression of Bcl-2. The apoptosis rate of SW480 cells treated by berberine was decreased, the expression of Bax was down-regulated, and the expression of Bcl-2 was up-regulated after BiX activation of GRP78. However, GRP94 expression did not change significantly in different treatment groups. Conclusion: Berberine can promote the apoptosis of SW480 cells by down-regulating GRP78 expression.
