从体内抑制试验和体外培养巨噬细胞的杀瘤试验对甲鱼组织浆的抑瘤作用进行了研究。体内抑瘤试验采用组织浆灌胃方式 ,计算抑瘤率和生命延长率。体外试验将巨噬细胞与肿瘤细胞共培养 ,测定NO浓度与细胞毒效应。培养上清液中一氧化氮 (NO)浓度采用NO试剂盒检测 ,细胞毒效应通过MTT比色法检测。用黄嘌呤氧化酶法测定服用鳖组织浆大鼠脑、肝、血中的超氧化物歧化酶 (SOD)含量。结果表明 ,不同来源鳖的效果有区别 ,野生鳖和仿生养殖鳖 2龄与 4龄对S180 小鼠肿瘤生长有明显的抑制作用 (P <0 .0 1) ,其抑瘤率分别达 70 .6 9%和 6 3.17% ,荷瘤小鼠的生命延长率均显著高于空白对照组 (P <0 .0 1) ,分别为40 .5 9%和 39.95 % ;1龄与 2龄温室鳖的抑瘤效果也显著 (P <0 .0 5 )。不同来源和不同年龄的鳖抑瘤效果有差异 ,野生鳖、仿生鳖和温室鳖组织浆能有效地增强巨噬细胞的细胞毒活性和NO水平 (P <0 .0 5~ 0 .0 1) ,且随年龄增长 ,效果增加。说明 ,鳖组织浆可活化巨噬细胞 ,诱生NO的产生 ,增加杀瘤效应。鳖的组织浆能提高大鼠脑、肝、血中的SOD活性 ,这种增强机体的抗衰老能力 ,随鳖的生活环境而有变化
The anti-tumor effects of turtle tissue plasm from in vivo inhibition test and in vitro killing experiment of macrophages were studied. Tumor inhibition test in vivo using tissue gavage method to calculate the tumor inhibition rate and life extension rate. In vitro experiments, macrophages were co-cultured with tumor cells to determine NO concentration and cytotoxic effect. The concentration of nitric oxide (NO) in culture supernatant was detected by NO kit, and the cytotoxic effect was detected by MTT colorimetric assay. The content of superoxide dismutase (SOD) in the brain, liver and blood of turtle tissue rats was measured by xanthine oxidase method. The results showed that the effect of different sources of turtle has a difference, the wild turtle and bionic turtle 2 and 4 instar S180 mouse tumor growth was significantly inhibited (P <0.01), the inhibition rate of 70. 6 9% and 6 3.17% respectively. The life prolongation in tumor-bearing mice was significantly higher than that in the blank control group (P <0.01), which were 40.59% and 39.95% The antitumor effect was also significant (P <0.05). The antitumor effects of turtle from different origins and ages were different. Tissue slurry of wild turtle, bionic turtle and greenhouse soft-shelled turtle could effectively enhance the cytotoxic activity and NO level of macrophages (P <0.05-0.01) , And with age, the effect increases.
