目的 研究肝细胞肝癌组织中PTEN与hTERT的表达及其意义 ,并探讨两者在肝细胞肝癌中表达的相关性。方法 应用免疫组织化学 (SP法 )和半定量逆转录 聚合酶链反应 (RT PCR)分析 5 8例肝细胞肝癌及对应癌旁组织中PTEN蛋白及PTENmRNA、hTERT蛋白及hTERTmRNA的表达情况 ,并探讨两者表达的相关性。结果 PTEN蛋白明确定位于肝细胞胞浆内。PTENmRNA和PTEN蛋白在肝细胞肝癌中的阳性率分别为 41.3 8% (2 4/ 5 8)、44 .83 % (2 6/ 5 8) ,明显低于癌旁肝组织的阳性率 10 0 % (5 8/ 5 8) ,差异有统计学意义 (P <0 .0 1)。hTERT蛋白主要表达于肝细胞胞浆内 ,偶见核内表达。hTERTmRNA和hTERT蛋白在肝细胞肝癌中的阳性率分别为82 .76% (4 8/ 5 8)和 87.93 % (5 1/ 5 8) ,明显高于癌旁组织中的阳性率 2 4.14 % (14 / 5 8)、2 7.5 9% (16/5 8) ,差异有统计学意义 (P <0 .0 1)。相关分析显示 ,PTEN与hTERT在肝细胞肝癌中的表达呈负相关 (P <0 .0 5 )。结论 PTEN、hTERT在肝细胞肝癌的发生发展中起重要作用 ,有可能成为肝细胞肝癌的重要分子生物学检测指标 ,且两者表达呈负相关 ,提示PTEN可能在肝细胞肝癌的发生发展过程中对hTERT的活化起着调控作用。
Objective To investigate the expression and significance of PTEN and hTERT in hepatocellular carcinoma (HCC) and to explore the correlation between the expression of PTEN and hTERT in hepatocellular carcinoma. Methods The expressions of PTEN, hTERT protein and hTERT mRNA in 58 hepatocellular carcinomas and corresponding para-cancerous tissues were analyzed by immunohistochemistry (SP method) and semi-quantitative reverse transcriptase-polymerase chain reaction (RT PCR) Correlation between the two. Results PTEN protein was localized in the cytoplasm of hepatocytes. The positive rates of PTEN mRNA and PTEN protein in hepatocellular carcinoma were 41.3 8% (44/83) and 44.83% (26/58), respectively, which were significantly lower than those in adjacent noncancerous liver tissues (100% (58/58), the difference was statistically significant (P <0.01). hTERT protein is mainly expressed in the cytoplasm of hepatocytes, occasionally nuclear expression. The positive rates of hTERT mRNA and hTERT protein in hepatocellular carcinoma were 82.76% (48/58) and 87.93% (51/58), respectively, which were significantly higher than those in paracancerous tissues (2.14% 14/58) and 2 7.59% (16/5 8), respectively. The difference was statistically significant (P <0.01). Correlation analysis showed that the expression of PTEN and hTERT in hepatocellular carcinoma was negatively correlated (P <0.05). Conclusions PTEN and hTERT play an important role in the development of hepatocellular carcinoma and may be an important molecular biological marker of hepatocellular carcinoma. The expression of PTEN and hTERT is negatively correlated, suggesting that PTEN may play an important role in the development of hepatocellular carcinoma Activation of hTERT plays a regulatory role.
