目的 采取手术标本 ,在裸鼠皮下连续传代建立人肾母细胞瘤裸鼠移植瘤模型。方法 切取肾母细胞瘤患儿肿瘤组织植入裸小鼠右前肢皮下 ,连续传代。观测肿瘤生长 ,对各代移植瘤进行光镜和电镜观察 ,波形蛋白 (vimentin)及上皮膜抗原 (EMA)免疫组化染色 ,WT1基因检测。结果 10个月中连续在裸鼠皮下传 4代 ,总成瘤率为 43%。移植瘤生长曲线与Gompentz函数符合。移植瘤保留了原肾母细胞瘤的组织学特征。Vimentin在各代肿瘤细胞中均可见表达。而EMA在本肿瘤中均无阳性表现。图像分析结果提示各代移植瘤DNA指数及其细胞时相分布基本一致。WT1基因3段短重复序列长度在各代之间无变化。结论 该模型基本保留了原肿瘤的生物学特性 ,是肾母细胞瘤基础及临床研究的良好模型。
Objective To take a surgical specimen and establish a human renaloblastoma xenograft tumor model in nude mice subcutaneously. Methods The tumor tissues of nephroblastoma were implanted subcutaneously and serially in the right forelimb of nude mice. The growth of the tumor was observed. The xenografts were observed under light microscope and electron microscope. The immunohistochemical staining of vimentin and epithelial membrane antigen (EMA) and WT1 gene were performed. Results 10 months in nude mice were subcutaneously transmitted 4 generations, the total tumor rate was 43%. The growth curve of the xenografts coincided with the Gompentz function. Xenografts retain the histological features of the original nephroblastoma. Vimentin is expressed in all generations of tumor cells. EMA had no positive expression in this tumor. The results of image analysis suggested that the DNA index and cell phase distribution of all the transplanted tumors were basically the same. WT1 gene 3 short repeat length did not change between generations. Conclusion This model basically retains the biological characteristics of the original tumor and is a good model for basic and clinical research of nephroblastoma.
