目的 揭示巨噬细胞集落刺激因 子(M-CSF)对单核/巨噬细胞氧化损伤的影响。方法采用真核细胞基因导入 的脂质体转染方法,建立了两株M-CSF基因过表达的RAW264.7细胞系,并观察 了氧化剂叔丁基氢过氧化物(tbOOH)对该RAW264.7细胞的氧化损伤作用。结果用M-CSF基因转染的RAW264.7细胞具 有较强的分泌有活性M-CSF的能力,而与正常RAW264.7细胞相比,M-CSF 过表达的RAW264.7细胞更易受到氧化剂的损伤。结论在一定条件下,M-CSF可促进巨噬细胞的氧化损伤。
Objective To investigate the effect of macrophage colony-stimulating factor (M-CSF) on the oxidative damage of monocytes / macrophages. Methods Two RAW264.7 cell lines overexpressing M-CSF gene were constructed by liposome transfection method using eukaryotic gene transfection. The effect of oxidant t-butyl hydroperoxide (tbOOH) on the growth of RAW264.7 cells The role of oxidative damage. As a result, RAW264.7 cells transfected with the M-CSF gene had a strong ability to secrete active M-CSF, whereas RAW264.7 cells overexpressing M-CSF were more susceptible to oxidative stress than did RAW264.7 cells damage. Conclusion Under certain conditions, M-CSF can promote macrophage oxidative damage.
