目的:观察加味四逆散药物血清对瘦素刺激肝星状细胞增殖的影响,探讨其抗肝纤维化的机制。方法:实验动物灌胃加味四逆散煎剂7d,制备各药物含药血清,并将药物血清用培养基配制成5%、10%、20%、40%四个浓度;培养肝星状细胞,取对数生长期细胞,使用噻唑兰(MTT)比色法检测药物血清对100ng/ml的瘦素刺激HSC-T6增殖的影响。结果:结果显示加味四逆散和秋水仙碱药物血清均有抑制肝星状细胞增殖的作用(P<0.05),且在5%~40%的血清浓度范内,两组对抑制HSC-T6细胞增殖抑制作用呈剂量依赖性的关系,但在10%~40%的血清浓度范围内对HSC-T6细胞增殖抑制率加味四逆散组高于秋水仙碱组(P<0.05)。结论:加味四逆散药物血清具有抑制瘦素刺激HSC-T6增殖的作用。
Objective: To observe the effects of Jiawei Sini Powder on the proliferation of hepatic stellate cells stimulated by leptin and to explore its mechanism of anti-liver fibrosis. Methods: The experimental animals were treated with Jiawei Sini Decoction for 7 days, and the drug-containing serums of each drug were prepared. The drug serum was formulated into four concentrations of 5%, 10%, 20% and 40% The cells in logarithmic growth phase were harvested and the effect of serum on the proliferation of HSC-T6 stimulated by leptin at 100ng / ml was determined by MTT assay. Results: The results showed that both Jiawei Sini San and colchicine serum inhibited the proliferation of hepatic stellate cells (P <0.05), and within the range of 5% ~ 40% of the serum concentration, the two groups inhibited HSC-T6 The inhibition of cell proliferation in a dose-dependent manner, but in the serum concentration range of 10% to 40% HSC-T6 cell proliferation inhibition rate of Sini powder group was higher than the colchicine group (P <0.05). Conclusion: Modified Sini Powder serum can inhibit the proliferation of HSC-T6 stimulated by leptin.
