目的:从野菊花中分离纯化多糖,并对其进行相对分子质量测定及单糖组成分析。方法:经热水提取、乙醇沉淀、大孔吸附树脂LSA-21脱色与Sevag法除蛋白,再经DEAE-52纤维素柱和Sephadex G75凝胶色谱分离纯化得到野菊花多糖。采用高效凝胶过滤色谱法(HPGPC)分析其相对分子质量,并借助衍生化气相色谱法(GC)对其单糖组成进行解析。结果:从野菊花中得到3个均一多糖CIP-1’,CIP-2’和CIP-3’。结论:CIP-1’的相对分子质量为8 242 Da,由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖6种单糖组成,摩尔比为0.75∶1.48∶1.96∶0.72∶1.80∶3.00;CIP-2’的相对分子质量为8 383 Da,由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖6种单糖组成,摩尔比为1.17∶1.19∶0.23∶0.56∶1.70∶2.20;CIP-3’的相对分子质量为19 201 Da,由鼠李糖、阿拉伯糖、甘露糖、葡萄糖和半乳糖5种单糖组成,摩尔比为2.40∶1.43∶3.84∶7.45∶5.10。红外光谱表明,三者均为吡喃糖。
OBJECTIVE: To isolate and purify polysaccharides from wild chrysanthemum, and to determine their relative molecular mass and monosaccharide composition. Methods: The polysaccharides of wild chrysanthemum were isolated and purified by hot water extraction, ethanol precipitation, macroporous adsorption resin LSA-21 decolorization and Sevag method, then by DEAE-52 cellulose column and Sephadex G75 gel chromatography. The relative molecular mass was analyzed by high performance gel permeation chromatography (HPGPC) and its monosaccharide composition was analyzed by derivatization gas chromatography (GC). Results: Three homogeneous polysaccharides CIP-1 ’, CIP-2’ and CIP-3 ’were obtained from wild chrysanthemum. CONCLUSION: The relative molecular mass of CIP-1 ’is 8 242 Da, consisting of 6 monosaccharides of rhamnose, arabinose, xylose, mannose, glucose and galactose in molar ratios of 0.75: 1.48: 1.96: 0.72: 1.80: 3.00; CIP-2 ’has a relative molecular mass of 8 383 Da and consists of 6 monosaccharides of rhamnose, arabinose, xylose, mannose, glucose and galactose in a molar ratio of 1.17: 1.19: 0.23: 0.56: 1.70: 2.20; CIP-3 ’has a relative molecular mass of 19-201 Da and consists of 5 monosaccharides of rhamnose, arabinose, mannose, glucose and galactose with the molar ratio of 2.40: 1.43: 3.84: 7.45 : 5.10. Infrared spectroscopy showed that all three were pyranose.
