目的:观察阿司匹林对人结肠癌肿瘤干细胞标志物CD44的影响,以探讨其对结肠癌干细胞的作用。方法:Western blot检测不同浓度阿司匹林作用后CD44在人结肠癌细胞株HT-29和SW480中的表达情况。结果:Western blot结果显示肿瘤干细胞标志物CD44在人结肠癌细胞株HT-29和SW480中均有表达,阿司匹林能降低HT-29和SW480中CD44蛋白的表达,并具有良好的量-效关系(P<0.05);两种细胞中CD44蛋白的表达比较差异均无统计学意义(P>0.05)。结论:阿司匹林能降低结肠癌细胞株HT-29和SW480中CD44的表达,并对结肠癌肿瘤干细胞有抑制作用。
Objective: To observe the effect of aspirin on human colon cancer stem cell marker CD44 to explore its effect on colon cancer stem cells. Methods: Western blot was used to detect the expression of CD44 in human colon cancer cell lines HT-29 and SW480 after different concentrations of aspirin. Results: The results of Western blot showed that CD44 was expressed in human colon cancer cell lines HT-29 and SW480. Aspirin could reduce the expression of CD44 protein in HT-29 and SW480 and had a good dose-response relationship P <0.05). There was no significant difference in the expression of CD44 between the two cell types (P> 0.05). Conclusion: Aspirin can reduce the expression of CD44 in colon cancer cell lines HT-29 and SW480, and inhibit the growth of colon cancer stem cells.
