目的 研究脑出血(ICH)后血肿周围区细胞凋亡的时间规律及胞磷胆碱钠治疗对血肿大小、细胞凋亡的影响。方法 自体非抗凝动脉血注入大鼠右侧尾壳核制作脑内出血模型;腹腔注射胞磷胆碱钠或生理盐水;按多田公式估算血肿体积;流式细胞仪检测血肿周围区细胞凋亡率。结果 大鼠脑内出血后6 h 血肿周围区出现细胞凋亡,72 h达高峰,4周时有少量凋亡细胞存在;对应的相同时间点给药组与对照组大鼠脑内血肿体积比较无显著差异(P>0.05);除6 h外余各时间点均显示给药组细胞凋亡率低于对照组,尤以72 h组为甚(P<0.01)。结论 脑内出血后血肿周围脑组织发生细胞凋亡且有时间规律性;胞磷胆碱钠早期治疗实验性大鼠脑内出血不引起血肿扩大,且可减少血肿周围区细胞凋亡。
Objective To study the time pattern of apoptosis in the perihematomal area after intracerebral hemorrhage (ICH) and the effect of citicoline on hematoma size and apoptosis. Methods Autologous non-anticoagulated arterial blood was injected into caudate putamen nucleus to make intracerebral hemorrhage model. Intraperitoneal injection of citicoline or saline was used. The volume of hematoma was estimated according to Tada formula. Flow cytometry was used to detect the apoptosis rate . Results Apoptosis occurred in the perihematomal area of the hematoma at 6 h after intracerebral hemorrhage in rats and peaked at 72 h after the intracerebral hemorrhage. A small amount of apoptotic cells existed at 4 weeks. Compared with the intracerebral hematoma volume at the same time points (P> 0.05). The apoptotic rates of all the groups at 6 hours were lower than that of the control group (P <0.01). Conclusions The intracerebral hemorrhage can induce the apoptosis of the brain tissue around the hematoma and have time regularity. The early treatment of intracerebral hemorrhage with citicoline does not cause hematoma enlargement in the intracerebral hemorrhage, and can reduce the apoptosis in the perihematoma area.
